Diet-related effects of antimicrobials in aquatic decomposer-shredder and periphyton-grazer systems

Leaf-associated microbial decomposers as well as periphyton serve as important food sources for detritivorous and herbivorous macroinvertebrates (shredders and grazers) in streams. Shredders and grazers, in turn, provide not only collectors with food but also serve as prey for predators. Therefore, decomposer-shredder and periphyton-grazer systems (here summarized as freshwater biofilm-consumer systems) are highly important for the energy and nutrient supply in heterotrophic and autotrophic stream food webs. However, both systems can be affected by chemical stressors, amongst which antimicrobials (e.g., antibiotics, fungicides and algaecides) are of particular concern. Antimicrobials can impair shredders and grazers not only via waterborne exposure (waterborne effect pathway) but also through dietary exposure and microorganism-mediated alterations in the food quality of their diet (dietary effect pathway). Even though the relevance of the latter pathway received more attention in recent years, little is known about the mechanisms that are responsible for the observed effects in shredders and grazers. Therefore, the first objective of this thesis was to broaden the knowledge of indirect antimicrobial effects in a model shredder and grazer via the dietary pathway. Moreover, although freshwater biofilm-consumer systems are most likely exposed to antimicrobial mixtures comprised of different stressor groups, virtually nothing is known of these mixture effects in both systems. Therefore, the second objective was to assess and predict diet-related antimicrobial mixture effects in a model freshwater biofilm-consumer system. During this thesis, positive diet-related effects of a model antibiotic on the energy processing and physiology of the shredder Gammarus fossarum were observed. They were probably triggered by shifts in the leaf-associated microbial community in favor of aquatic fungi that increased the food quality of leaves for the shredder. Contrary to that, a model fungicide induced negative effects on the energy processing of G. fossarum via the dietary pathway, which can be explained by negative impacts on the microbial decomposition efficiency leading to a reduced food quality of leaf litter for gammarids. For diet-related antimicrobial effects in periphyton-grazer systems, a model algaecide altered the periphyton community composition by increasing nutritious and palatable algae. This resulted in an enhanced consumption and physiological fitness of the grazer Physella acuta. Finally, it was shown that complex horizontal interactions among leaf-associated microorganisms are involved, making diet-related antimicrobial mixture effects in the shredder G. fossarum difficult to predict. Thus, this thesis provides new insights into indirect diet-related effects of antimicrobials on shredders and grazers as well as demonstrates uncertainties of antimicrobial mixture effect predictions for freshwater biofilm-consumer systems. Moreover, the findings in this thesis are not only informative for regulatory authorities, as indirect effects and effects of mixtures across chemical classes are not considered in the environmental risk assessment of chemical substances, but also stimulate future research to close knowledge gaps identified during this work.
To render the surface of a material capable of withstanding mechanical and electrochemical loads, and to perform well in service, the deposition of a thin film or coating is a solution. In this project, such a thin film deposition is carried out. The coating material chosen is titanium nitride (TiN) which is a ceramic material known to possess a high hardness (>10 GPa) as well as good corrosion resistance. The method of deposition selected is high power impulse magnetron sputtering (HiPIMS) that results in coatings with high quality and enhanced properties. Sputtering is a physical process that represents the removal or dislodgment of surface atoms by energetic particle bombardment. The term magnetron indicates that a magnetic field is utilized to increase the efficiency of the sputtering process. In HiPIMS, a high power is applied in pulses of low duty cycles to a cathode that is sputtered and that consists of the coating material. As result of the high power, the ionization of the sputtered material takes place giving the possibility to control these species with electric and magnetic field allowing thereby the improvement and tuning of coating properties. However, the drawback of HiPIMS is a low deposition rate.
In this project, it is demonstrated first that it is possible to deposit TiN coating using HiPIMS with an optimized deposition rate, by varying the magnetic field strength. It was found that low magnetic field strength (here 22mT) results in a deposition rate similar to that of conventional magnetron sputtering in which the average power is applied continuously, called also direct current magnetron sputtering (dcMS). The high deposition rate at low magnetic field strength was attributed to a reduction in the back attraction probability of the sputtered species. The magnetic field strength did not show noticeable influence on the mechanical properties. The proposed explanation was that the considered peak current density interval 1.22-1.72 A∙cm-2 does not exhibit dramatic changes in the plasma dynamics.
In a second part, using the optimized deposition rate, the optimized chemical composition of TiN was determined. It was shown that the chemical composition of TiN does not significantly influence the corrosion performance but impacts considerably the mechanical properties. It was also shown that the corrosion resistance of the coatings deposited using HiPIMS was higher than that of the coatings deposited using dcMS.
The third study was the effect of annealing post deposition on the properties of TiN coating deposited using HiPIMS. The hardness of the coatings showed a maximum at 400°C reaching 24.8 GPa. Above 400°C however, a lowering of the hardness was measured and was due to the oxidation of TiN which led to the formation of TiN-TiO2 composites with lower mechanical properties.
The coating microscopic properties such as crystal orientation, residual stresses, average grain size were determined from X-ray diffraction data and the roughness was measured using atomic force microscopy. These properties were found to vary with the magnetic field strength, the chemical composition as well as the annealing temperature.
Unternehmen versuchen, Knowledge Management (KM) zu nutzen, um mehr Effizienz und Effektivität im Unternehmen zu erreichen. Das Hauptproblem besteht darin, dass die meisten dieser KM Projekte nicht oder nur selten auf nachhaltigen Analysen oder etablierten Theorien über KM basieren. Oft besteht eine große Kluft zwischen den Erwartungen und dem tatsächlichen Ergebnis solcher KM Initiativen. Die zu beantwortende Forschungsfrage lautet also: Welche Herausforderungen ergeben sich in KM Projekten, welche KM Anforderungen können daraus abgeleitet werden und welche Empfehlungen unterstützen das Ziel, die Anforderungen an KM zu erfüllen? Als theoretische Grundlage wird eine Reihe von KM Frameworks untersucht. Anschließend werden KM Herausforderungen aus der Literatur analysiert und mit Best-Practice-Beispielen aus Case-Studies werden Handlungsempfehlungen für diese Herausforderungen gegeben. Das Hauptergebnis dieser Arbeit ist eine Best Practice-Guideline, die es Chief Knowledge Officers (CKOs) und KM Projektmanagern ermöglicht, die in dieser Arbeit genannten Herausforderungen genau zu untersuchen und eine geeignete Methode zu finden, diese Herausforderung optimal zu meistern. Diese Guideline zeigt, dass KM auf vielfältige Weise positiv und negativ beeinflusst werden kann. Knowledge Management (KM) in einem Unternehmen zu meistern ein großes und weitreichendes Unterfangen ist und das Technologie bzw. Information Technology (IT) nur ein Teil des großen Ganzen ist.
Das Internet der Dinge (IoT) ist ein Netzwerk bestehend aus adressierbaren, physikalischen Objekten, die Sensor-, Kommunikations- und Aktuator-Technologien bereitstellen und mit ihrer Umwelt interagieren (Geschickter 2015). Wie jedes neue Konzept, hat auch IoT Interesse über jeden Anwendungsbereich hinweg, sowohl in Theorie als auch Praxis, geweckt und die verfügbaren Technologien an ihre Grenzen gebracht. Diese Grenzen machen sich insbesondere dann bemerkbar, wenn die Anzahl von Dingen (Things), die über verschiedenste Anwendungsbereiche hinweg verwaltet werden müssen, steigt. Um die neuartigen Anforderungen zu erfüllen, wurde eine Fülle von verschiedenen Systemen entwickelt, die alle ihre eigenen Interpretationen einer IoT Architektur und ihrer jeweiligen Komponenten anwenden. Dies hat dazu geführt, dass IoT aktuell eher ein Intranet der Dinge als ein Internet der Dinge ist (Zorzi et al. 2010). Daher ist es Ziel dieser Arbeit, ein einheitliches Verständnis der Komponenten, die eine IoT Architektur bilden, zu erlangen und generische Spezifikationen in Form eines Ganzheitlichen IoT Architektur Frameworks zur Verfügung zu stellen.
Diese Arbeit verwendet Design Science Research (DSR), um die genannte Architektur auf Basis der einschlägigen Literatur zu entwickeln. Die Entwicklung des Ganzheitlichen IoT Architektur Frameworks umfasst die Nutzung zwei neuer Perspektiven auf IoT Architekturen (IoT Architecture Perspectives), die während der Analyse von IoT Architekturen in der Literatur identifiziert wurden. Die Anwendung dieser neuen Perspektiven führte zur Erkenntnis, dass eine weitere, ebenfalls neuartige, Komponente in der Literatur implizit erwähnt wird. Die Beschreibungen der Komponenten von verschiedenen IoT Architekturen wurden vereinheitlicht und mit der neuen Komponente, dem Thing Management System, in Beziehung gesetzt, um das Ganzheitliche IoT Architektur Framework zu entwickeln. Weiterhin wurde gezeigt, dass die Spezifikationen der Architektur als Vorlage für die Implementation eines Prototypen geeignet ist.
Der Hauptbeitrag dieser Arbeit ist ein vereinheitlichtes Verständnis der einzelnen Komponenten sowie deren Interaktionen einer IoT Architektur.
The development of a game engine is considered a non-trivial problem. [3] The architecture of such simulation software must be able to manage large amounts of simulation objects in real-time while dealing with “crosscutting concerns” [3,p. 36] between subsystems. The use of object oriented paradigms to model simulation objects in class hierarchies has been reported as incompatible with constantly changing demands during game development [2, p. 9], resulting in anti-patterns and eventual, messy refactoring.[13]
Alternative architectures using data oriented paradigms revolving around object composition and aggregation have been proposed as a result. [13, 9, 1, 11]
This thesis describes the development of such an architecture with the explicit goals to be simple, inherently compatible with data oriented design, and to make reasoning about performance characteristics possible. Concepts are formally defined to help analyze the problem and evaluate results. A functional implementation of the architecture is presented together with use cases common to simulation software.
The estimation of various social objects is necessary in different fields of social life, science, education, etc. This estimation is usually used for forecasting, for evaluating of different properties and for other goals in complex man-machine systems. At present this estimation is possible by means of computer and mathematical simulation methods which is connected with significant difficulties, such as: - time-distributed process of receiving information about the object; - determination of a corresponding mathematical device and structure identification of the mathematical model; - approximation of the mathematical model to real data, generalization and parametric identification of the mathematical model; - identification of the structure of the links of the real social object. The solution of these problems is impossible without a special intellectual information system which combines different processes and allows predicting the behaviour of such an object. However, most existing information systems lead to the solution of only one special problem. From this point of view the development of a more general technology of designing such systems is very important. The technology of intellectual information system development for estimation and forecasting the professional ability of respondents in the sphere of education can be a concrete example of such a technology. Job orientation is necessary and topical in present economic conditions. It helps tornsolve the problem of expediency of investments to a certain sphere of education. Scientifically validated combined diagnostic methods of job orientation are necessary to carry out professional selection in higher education establishments. The requirements of a modern society are growing, with the earlier developed techniques being unable to correspond to them sufficiently. All these techniques lack an opportunity to account all necessary professional and personal characteristics. Therefore, it is necessary to use a system of various tests. Thus, the development of new methods of job orientation for entrants is necessary. The information model of the process of job orientation is necessary for this purpose. Therefore, it would be desirable to have an information system capable of giving recommendations concerning the choice of a trade on the basis of complex personal characteristics of entrants.
Wildschweine gehören zu den meist verbreiteten Huftieren der Welt. Die Wildschweinpopulation in Deutschland stieg in den letzten drei Jahrzehnten deutlich an. Heutzutage führt deren hohe Dichte zu Schäden auf landwirtschaftlichen Flächen und spielt eine wichtige Rolle bei der Ausbreitung von Krankheiten. Für ein effektives Wildschwein Management sind Informationen über absolute Populationszahlen von höchster Bedeutung. Es existieren verschiedene traditionelle Methoden wie z.B. die direkte Beobachtung der Tiere, das Zählen ihrer Losungen oder die Auswertung der Statistiken zu Jagderträgen, die nur relative Schätzungen oder Populationstrends liefern. Absolute Populationszahlen könnte der Fang-Markier-Wiederfang Ansatz hervorbringen. Nichtsdestotrotz ist das Fangen von Wildschweinen schwer zu realisieren und kostenaufwendig in Bezug auf die hierzu benötigten Arbeitskräfte und den Feldarbeitsaufwand.
Weiterhin sind die Fangwahrscheinlichkeiten heterogen aufgrund der Verhaltensvariabilitaet der Individuen, die durch Alter, Geschlecht und Erfahrung der Tiere bedingt ist. Nicht-invasive genetische Verfahren sind vielversprechender gegenüber den traditionellen Methoden der Populationsgrößenschätzung speziell für Wildschweine. Denn zum Einen reduzieren diese Methoden den Stressfaktor der Tiere und senken Fangfehler und zum Anderen erhöhen sie die Zahl indirekter Beobachtungen. Kot eignet sich sehr gut als DNS-Quelle für die Wildschwein Genotypisierung, da dieser eine nahezu gleiche Erfassungswahrscheinlichkeit für alle Tiere gewährleistet. Nichtsdestotrotz bringt das Arbeiten mit Kot Schwierigkeiten mit sich, die sich in in einer geringen DNS Qualität und Quantität oder Genotypisierungsfehlern äußern. Das Hauptziel der vorliegenden Arbeit war die Entwicklung einer zuverlässigen, kostengünstigen, reproduzierbaren und praktikablen Methode zur Genotypisierung von Wildschweinen. Diese Methode sollte einen zuverlässigen Genotypendatensatz liefern, der aus der Genotypisierung gesammelter Kotproben stammte. Individuelle Identifikation bildet die Basis für einen anzuwendenden Fang-Markierung-Wiederfang Ansatz. Da es bisher keine vergleichbaren Untersuchungen zu Populationsschätzungen von Wildscheinen gibt, fehlen Referenzdaten zum nicht-invasiven Ansatz. Daher wurden verschiedene Versuche zur Reduzierung und Quantifizierung der Genotypisierungsfehlerraten (GFR) getestet, verglichen und evaluiert. Um die Amplifizierungsrate zu erhöhen wurden Hälterungs- und DNS-Isolationsverfahre optimiert. Ein Schritt für Schritt Ansatz zur Bestimmung der minimal erforderlichen Anzahl von Mikrosatelliten Marker wurde entwickelt, welcher einen Test mit nahverwandten Individuen (Mütter und deren Föten) beinhaltete, um sogar diese voneinander zu unterscheiden. Ein so genanntes Multitube-Verfahren wurden angewendet um die GFR zu reduzieren. Die Quantifizierung von GFR aus erhobenen Datensätzen wurde evaluiert indem verschiedene Methoden zur GFR-Bestimmung getestet und miteinander verglichen wurden, hierbei ergaben sich GFR zwischen 0% bis 54%. Als Konsequenz dessen wurden die Kriterien für das Multitube-Verfahren verschärft, indem die Anzahl der Wiederholungen von homozygoten Proben erhöht wurde. Eine zusätzliche Validierung in Form eines Blindtests wurde etabliert, um die Zuverlässigkeit der Genotypisierung und Fehlerkorrekturen zu bekräftigen.
Abschließend wurde ein strikter und praktikabler Verfahrenvorschlag entwickelt, beginnend beim Sammeln der Kotproben und endend mit dem Erhalt eines zuverlässigen Datensatzes mit Genotypen einzelner Proben.
Die Ergebnisse der hier präsentierten Methode aus zwei Beprobungen 2006 und 2007 in einem 4000 ha großen Areal im Pfälzer Wald führte zu ungenauen Schätzungen mit hohen Konfidenzintervallen (KI). So lag die geschätzte Populationsgrösse in der Beprobung 2006 bei 215 Individuen, was 156-314 (KI 95%) Individuen entspricht. Die Populationsschätzung 2007 brachte 415 Individuen hervor, was 315-561 (KI 95%) Individuen entspricht. Dies ließ auf zu niedrige Stichproben (2006 betrug n = 141, 2007 n = 326), zu wenig erfolgreich analysierte Proben (2006 n = 89, 2007 n = 156) und/ oder zu wenig Wiederfänge (2006 n = 12, 2007 n = 24) schließen. Zudem ergaben die Schätzungen sogar deutlich höhere Populationszahlen als zuvor durch Jagderträge vermutet wurde, was auf eine uneffektive Bejagungsstrategie in dem Studienareal hindeutet. Für den zukünftigen Ausblick ist es unabdingbar die Stichprobenzahl deutlich zu erhöhen um die Validität und Reliabiltät der Populationsschätzungen zu gewährleisten, da diese für das Wildmanagement und die epidemiologischen Lösungsstrategien von höchster Bedeutung sind. Nichtsdestotrotz konnte die im Rahmen dieser Arbeit entwickelte Methode zur individuellen Wildschwein Genotypisierung erfolgreich etabliert werden. Die daraus resultierenden Datensets zur Modellierung von Populationschätzungen sind zuverlässig und weisen eine ausreichend geringe reale Genotypisierungsfehlerrate auf.
Leichte Sprache (LS) ist eine vereinfachte Varietät des Deutschen in der barrierefreie Texte für ein breites Spektrum von Menschen, einschließlich gering literalisierten Personen mit Lernschwierigkeiten, geistigen oder entwicklungsbedingten Behinderungen (IDD) und/oder komplexen Kommunikationsbedürfnissen (CCN), bereitgestellt werden. LS-Autor*innen sind i.d.R. der deutschen Standardsprache mächtig und gehören nicht der genannten Personengruppe an. Unser Ziel ist es, diese zu befähigen, selbst am schriftlichen Diskurs teilzunehmen. Hierfür bedarf es eines speziellen Schreibsystems, dessen linguistische Unterstützung und softwareergonomische Gestaltung den spezifischen Bedürfnissen der Zielgruppe gerecht wird. EasyTalk ist ein System basierend auf computerlinguistischer Verarbeitung natürlicher Sprache (NLP) für assistives Schreiben in einer erweiterten Variante von LS (ELS). Es stellt den Nutzenden ein personalisierbares Vokabular mit individualisierbaren Kommunikationssymbolen zur Verfügung und unterstützt sie entsprechend ihres persönlichen Fähigkeitslevels durch interaktive Benutzerführung beim Schreiben. Intuitive Formulierungen für linguistische Entscheidungen minimieren das erforderliche grammatikalische Wissen für die Erstellung korrekter und kohärenter komplexer Inhalte. Einfache Dialoge kommunizieren mit einem natürlichsprachlichen Paraphrasengenerator, der kontextsensitiv Vorschläge für Satzkomponenten und korrekt flektierte Wortformen bereitstellt. Außerdem regt EasyTalk die Nutzer*innen an, Textelemente hinzuzufügen, welche die Verständlichkeit des Textes für dessen Leserschaft fördern (z.B. Zeit- und Ortsangaben) und die Textkohärenz verbessern (z.B. explizite Diskurskonnektoren). Um das System auf die Bedürfnisse der Zielgruppe zuzuschneiden, folgte die Entwicklung von EasyTalk den Grundsätzen der menschzentrierten Gestaltung (UCD). Entsprechend wurde das System in iterativen Entwicklungszyklen ausgereift, kombiniert mit gezielten Evaluierungen bestimmter Aspekte durch Gruppen von Expert*innen aus den Bereichen CCN, LS und IT sowie L2-Lernende der deutschen Sprache. Eine Fallstudie, in welcher Mitglieder der Zielgruppe das freie Schreiben mit dem System testeten, bestätigte, dass Erwachsene mit geringen Lese-, Schreib- und Computerfähigkeiten mit IDD und/oder CCN mit EasyTalk eigene persönliche Texte in ELS verfassen können. Das positive Feedback aller Tests inspiriert Langzeitstudien mit EasyTalk und die Weiterentwicklung des prototypischen Systems, wie z.B. die Implementierung einer s.g. Schreibwerkstatt.
Prozedurale Synthese, das Erzeugen von Computerspielinhalten durch die Verwendung von pseudo-zufälligen Algorithmen, ist ein Themenbereich mit wachsendem Interesse in wirtschaftlichen und akademischen Kreisen, verdankt sowohl durch ihre Eignung zur Senkung von Entwicklungszeit und -kosten als auch durch die Möglichkeit, prozedurale Synthese zur Erzeugung von interessanten und einmaligen Spielwelten einzusetzen. Obwohl viele aktuelle Computerspiele prozedural generierte Inhalte verwenden, gebrauchen nur wenige diese Methoden zur Erzeugung realistischer Weltraum-Spielumgebungen, und die Umsetzbarkeit der Anwendung prozeduraler Synthese zur Entwicklung eines solchen Spiels wurde untersucht. Aktuelle Modelle aus der Forschung wurden verwendet, um eine in Echtzeit laufende astronomische Simulation in Python zu entwickeln, welche Stern- und Planetenobjekte in einer fiktiven Galaxie prozedural erzeugt, die als Spielwelt eines einfachen 2DWeltraumspiels dient, in welchem der Spieler nach intelligentem Leben suchen muss.
Die Magnetresonanztomographie (MRT) ist ein bildgebendes Verfahren, das in der medizinischen Diagnostik zur Darstellung von Struktur und Funktion der Gewebe und Organe im Körper eingesetzt wird. Diffusionsgewichtete Bildgebung ist ein spezielles bildgebendes MRT Verfahren, welches es ermöglicht, nichtinvasiv und in vivo Einblicke in den Verlauf von Nervenbahnen zu geben. Es erlaubt damit, Aussagen über die Struktur und Integrität dieser Verbindungsbahnen zu treffen. Im klinischen Alltag findet diese Modalität Anwendung in der neurochirurgischen Operationsplanung, wie beispielsweise bei Resektionen von Läsionen, die in wichtigen funktionellen oder tiefiegenden Arealen liegen, wo die Beschädigungsgefahr wichtiger Nervenbahnen gegeben ist. Kommt es im Zuge der Operation zu einer etwaigen Durchtrennung von wichtigen Bahnen, kann dies zu erheblichen funktionellen Beeinträchtigung führen. Diese Arbeit gibt eine Einführung in die MRT-Bildgebung und wird sich im Speziellen mit der Aufnahme von diffusionsgewichtetenMRT- Daten beschäftigen. Generell besteht das Problem, dass das Auflösungsvermögen von Diffusionsdaten relativ niedrig ist in Relation zum Aufnahmeobjekt. So werden in einem einzelnen 3D Volumenelement, auch Voxel genannt, eine Reihe von Nerventrakten abgebildet, die sich beispielsweise kreuzen, aufsplitten oder auffächern. Hier besteht die Notwendigkeit, diese Voxel zu identifizieren und zu klassifizieren, um auch in schwierigen Regionen aus den lokalen Diffusionsdaten die Verläufe von Nervenbündeln möglichst exakt zu rekonstruieren. Diese Rekonstruktion wird durch die sogenannte Traktographie realisiert. Im Zuge dieser Arbeit werden wir existierende Rekonstruktionsmethoden, wie beispielsweise diffusion tensor imaging (DTI) und q-ball imaging (QBI) auf synthetisch generierten Daten untersuchen. Wir werden herausstellen, welche wertvollen Informationen die rekonstruierten Daten liefern können und welche individuellen Einschränkungen es gibt. QBI rekonstruiert eine orientation distribution function (ODF), deren lokalen Maxima in vielen Fällen mit den Richtungen der Nervenbahnen übereinstimmen. Wir bestimmen diese lokalen Maxima. Auf den Metriken des Diffusionstensors wird eine neue voxelbasierte Klassifikation vorgestellt. Die Vereinigung von voxelbasierter Klassifikation, lokalen Maxima und globalen Informationen aus der Nachbarschaft eines Voxels ist der Hauptbeitrag dieser Arbeit und führt zur Entwicklung eines globalen Klassifikators, der mögliche Traktographie-Richtungen vorgibt und asymmetrische Konfigurationen ermittelt. Im Anschluss wird ein eigener Traktographie-Algorithmus vorgestellt, der auf den Ergebnissen des globalen Klassifikators arbeitet und somit auch Aufsplittungen von Nervenbahnen abbilden kann.
Das World Wide Web (WWW) ist heute zu einem sehr wichtigen Kommunikationskanal geworden, dessen Nutzung in der Vergangenheit stetig gestiegen ist. Websitebesitzer haben schon seit der Entwicklung des ersten Web Browser von Tim Berners-Lee im Jahre 1990 Interesse daran, das Verhalten von Benutzern zu erkennen und zu verstehen. Durch den Einfluss, den der Onlinekanal heute erzielt und welcher alle anderen Medien übersteigt, ist auch das Interesse im Beobachten von Website-Benutzungen und Benutzeraktivitäten noch weiter gestiegen. Das Sammeln und Analysieren von Daten über die Benutzung von Websites kann helfen, Benutzerverhalten zu verstehen, Services zu verbessern und Gewinn zu steigern.
Darüber hinaus ist es Voraussetzung für effektives Website Design und Management, effektive Mass Customization und effektives Marketing. Das Themengebiet, welches diese Aspekte betrachtet, heißt Web Analytics (WA).rnAllerdings führen sich ändernde Technologien und sich entwickelnde Web Analytics Methoden und Prozesse zu großen Herausforderungen für Unternehmen, die Web Analytic Programme durchführen. Aufgrund fehlender Ressourcen in den verschiedensten Bereichen, sowie einer hier oft aufzufindenden anderen Art von Websites, ist es vor allem für Klein- und Mittelständige Unternehmen (KMU) sowie Non-Profit Organisationen schwer, Web Analytics in einer effektiven Weise zu betreiben.
Dieses Forschungsvorhaben zielt daher darauf ab, die vorhandene Lücke zwischen der Theorie, den Möglichkeiten die Tools bieten und den betrieblichen Anforderungen zu identifizieren. Hierfür wird das Thema von drei verschiedenen, jedoch aufeinander aufbauenden Richtungen betrachtet: der akademischen Literatur, Web Analytic Programmen und einer Fallstudie. Mit Hilfe eines Action Research Ansatzes war es möglich, einen ganzheitlichen Überblick des Themengebiets Web Analytics zu erhalten und bestehende Lücken aufzudecken. Das Ergebnis dieser wissenschaftlichen Arbeit ist ein Framework, welches KMUs die Informations-Websites betreiben dabei hilft, Web Analytic Initiativen durch zu führen.
Die automatische Identifikation von Experten in einer speziellen technologischen Domäne, wie einer Bibliothek, Framework oder generellen Technologie, schafft einen großen Mehrwert in der gemeinsamen Entwicklung von Softwareprojekten. Daher soll in dieser Arbeit ein Vorgehen sowie ein Programm zur automatischen Identifikation von Experten entwickelt werden, die gewissen Skills besitzen. Hierbei wird speziell das Django-Framework betrachtet. Jedoch kann durch hinzufügen von weiteren Regeln unser Tool leicht auf andere Technologien angepasst werden. Abschließend wird eine case study auf ein Open Source Projekt durchgeführt.
Designing Core Ontologies

One of the key factors that hinders integration of distributed, heterogeneous information systems is the lack of a formal basis for modeling the complex, structured knowledge that is to be exchanged. To alleviate this situation, we present an approach based on core ontologies. Core ontologies are characterized by a high degree of axiomatization and formal precision. This is achieved by basing on a foundational ontology. In addition, core ontologies should follow a pattern-oriented design approach. By this, they are modular and extensible. Core ontologies allow for reusing the structured knowledge they define as well as integrating existing domainrnknowledge. The structured knowledge of the core ontologies is clearly separated from the domain-specific knowledge. Such core ontologies allow for both formally conceptualize their particular fields and to be flexibly combined to cover the needsrnof concrete, complex application domains. Over the last years, we have developed three independent core ontologies for events and objects, multimedia annotations, and personal information management. In this paper, we present the simultaneousrnuse and integration of our core ontologies at the example of a complex, distributed socio-technical system of emergency response. We describe our design approach for core ontologies and discuss the lessons learned in designing them. Finally, we elaborate on the beauty aspects of our core ontologies.
Ziel dieser Arbeit ist es, ein Recommender System (RS) für Geschäftsprozesse zu erstellen, das auf dem bestehenden ProM-Plug-in RegPFA aufbaut. Um dies zu gewährleisten, soll zunächst eine Schnittstelle geschaffen werden, welche die von RegPFA erstellten probabilistischen endlichen Automaten (PFA) im tsml-Format zu einer erweiterbaren Datenbasis zusammenfassen kann. Anschließend soll ein Java-Programm geschrieben werden, das mithilfe dieser Datenbasis zu einem gegebenen Teilprozess die wahrscheinlichsten Empfehlungen für das nächstfolgende Prozesselement angibt.
Software Projekte nutzen typischerweise mehrere externe Programmbibliotheken. Die Schnittstelle, die solch eine Programmbibliothek zur Verfügung stellt, wird als API (application programming interface) bezeichnet. APIs werden üblicherweise laufend weiterentwickelt, was es notwendig macht, dass die Anwendungen, welche sie verwenden, entsprechend modifiziert werden. Zudem kann es kann vorkommen, dass eine Programmbibliothek durch eine andere ersetzt werden soll, was ebenfalls zur Folge hat, dass die Anwendungen, wo die API verwendet wurde, modifiziert werden müssen. Den Vorgang eine Anwendung so zu modifizieren, dass eine andere API verwendet wird, bezeichnet man als API Migration. Manuelle API Migration ist eine mühselige und zeitintensive Aufgabe, deshalb ist automatische API Migration ein aktives Forschungsfeld. Ein verwandtes Forschungsgebiet ist API Analyse, welches Daten zur Verfügung stellt, die helfen können Werkzeuge für API Migration zu entwickeln. Die hier vorliegende Arbeit behandelt Techniken und Technologien für die Entwicklung von Werkzeugen für API Analyse und API Migration. Die Ergebnisse werden als Design Patterns präsentiert, welche auf unseren Erfahrungen mit API Analyse und API Migration innerhalb des Software Languages Teams basieren.
Paketmanager für Quellcode wie zum Beispiel Cabal verwalten unter anderem die Abhängigkeiten zwischen Paketen. Softwareprojekte nutzen jedoch selten sämtliche Funktionalitäten, die ihre Abhängigkeiten bereitstellen. Das führt zur unnötigen Kompilation unbenutzter Code-Fragmente und zu vermeintlichen Versionskonflikten, wo gar keine Konflikte sind. In zwei Fallstudien zeigen wir, wie relevant diese zwei Probleme sind. Danach beschreiben wir, wie wir sie vermeiden können, indem wir Abhängigkeiten nicht zwischen ganzen Paketen, sondern zwischen einzelnen Code-Fragmenten feststellen.
Eine genaue Schneesimulation ist der Schlüssel zur Erfassung der charakteristischen Visualisierung von Schnee. Aufwendige Methoden existieren, die versuchen Schneeverhalten ganzheitlich zu ergreifen. Die Rechenkomplexität dieser Ansätze hindert sie daran, Echtzeitfähigkeit zu erreichen. Diese Arbeit stellt drei Methoden vor, die unter Verwendung der GPU eine echtzeitfähige Deformation einer Schneeoberoberfläche darstellen. Die Ansätze werden hinsichtlich ihrer wahrheitsgetreuen Schneedarstellung untersucht und nach ihrer Fähigkeit, mit einer zunehmenden Anzahl von schneeverformenden Objekten zu skalieren. Die Ergebnisse zeigen, dass die Methoden bei mehreren hunderten schneeverformenden Objekten ihre Echtzeitfähigkeit beibehalten. Jedoch benachteiligen die charakteristischen Einschränkungen jener Methoden die visuellen Resultate. Ein experimenteller Ansatz ist es, die Anzahl der schneeverformenden Objekte zu reduzieren und durch Zusammenfügen der Methoden ein genaueres, kombiniertes Verformungsmuster zu erzeugen.
Konstituenten-Parsing versucht, syntaktische Struktur aus einem Satz zu extrahieren. Diese Parsing-Systeme sind in vielen maschinellen Sprachverarbeitungsanwendungen hilfreich, wie z.B. bei der Grammatikprüfung, der Beantwortung von Fragen und der Informationsextraktion. In dieser Masterarbeit geht es um die Implementierung eines Konstituentenparsers für die deutsche Sprache mit Hilfe von neuronalen Netzen. In der Vergangenheit wurden wiederkehrende neuronale Netze beim Aufbau eines Parsers und auch bei vielen maschinellen Sprachverarbeitungsanwendungen verwendet. Dabei werden Module des neuronalen Netzes mit Selbstaufmerksamkeit intensivgenutzt, um Sätze effektiv zu verstehen. Bei mehrschichtigen Selbstaufmerksamkeitsnetzwerken erreicht das konstituierende
Parsen 93,68% F1-Scoret. Dies wird noch weiter verbessert, indem sowohl Zeichen- als auch Worteinbettungen als Darstellung des Inputs verwendet werden. Ein F1-Score von 94,10% wurde am besten durch den Konstituenten-Parser erreicht, der nur den bereitgestellten Datensatz verwendet. Mit Hilfe externer Datensätze wie der deutschen Wikipedia werden vortrainierte ELMo-Modelle zusammen mit Selbstbeobachtungsnetzwerken verwendet, die einen F1-Score von 95,87% erreichen.
Objectives: Crayfish plague disease, caused by the oomycete pathogen Aphanomyces astaci represents one of the greatest risks for the biodiversity of the freshwater crayfish. This data article covers the de novo transcriptome assembly and annotation data of the noble crayfish and the marbled crayfish challenged with Ap. astaci. Following the controlled infection experiment (Francesconi et al. in Front Ecol Evol, 2021, https://doi.org/10.3389/fevo.2021.647037), we conducted a differential gene expression analysis described in (Boštjančić et al. in BMC Genom, 2022, https://doi.org/10.1186/s12864-022-08571-z) Data description: In total, 25 noble crayfish and 30 marbled crayfish were selected. Hepatopancreas tissue was isolated, followed by RNA sequencing using the Illumina NovaSeq 6000 platform. Raw data was checked for quality with FastQC, adapter and quality trimming were conducted using Trimmomatic followed by de novo assembly with Trinity. Assembly quality was assessed with BUSCO, at 93.30% and 93.98% completeness for the noble crayfish and the marbled crayfish, respectively. Transcripts were annotated using the Dammit! pipeline and assigned to KEGG pathways. Respective transcriptome and raw datasets may be reused as the reference transcriptome assemblies for future expression studies.
Modern Internet and Intranet techniques, such as Web services and virtualization, facilitate the distributed processing of data providing improved flexibility. The gain in flexibility also incurs disadvantages. Integrated workflows forward and distribute data between departments and across organizations. The data may be affected by privacy laws, contracts, or intellectual property rights. Under such circumstances of flexible cooperations between organizations, accounting for the processing of data and restricting actions performed on the data may be legally and contractually required. In the Internet and Intranet, monitoring mechanisms provide means for observing and auditing the processing of data, while policy languages constitute a mechanism for specifying restrictions and obligations.
In this thesis, we present our contributions to these fields by providing improvements for auditing and restricting the data processing in distributed environments. We define formal qualities of auditing methods used in distributed environments. Based on these qualities, we provide a novel monitoring solution supporting a data-centric view on the distributed data processing. We present a solution for provenance-aware policies and a formal specification of obligations offering a procedure to decide whether obligatory processing steps can be met in the future.
